兔血管性血友病因子/瑞斯托霉素輔因子(VWF)ELISA試劑盒原理的簡要說明書：VWF(Von Willebrand Factor 血管性血友病因子)Von Willebrand Factor因子是一多聚糖蛋白，它與*VIII以非共價(jià)方式結(jié)合成復(fù)合物存在于血漿中。正常的止血過程中，高分子量多聚體形式的vWF可作為分子橋介導(dǎo)血小板球蛋白IB與內(nèi)皮下膠原的黏附反應(yīng)。先天性VWF異?？梢鹬卸戎林囟瘸鲅獌A向—此為Von Willebrand病(VWD)。提供的ELISA Kit是典型的夾心法酶聯(lián)免疫吸附測定試劑盒(ELISA)。預(yù)先包被的抗體為多克隆抗體。檢測相抗體也為多克隆抗體，經(jīng)*(biotin)標(biāo)記。樣品和*標(biāo)記抗體先后加入酶標(biāo)板孔反應(yīng)后，PBS或TBS洗滌。隨后加入過氧化物酶標(biāo)記的親和素反應(yīng)；經(jīng)過PBS或TBS的*洗滌后用底物TMB顯色。TMB在過氧化物酶的催化下轉(zhuǎn)化成藍(lán)色，并在酸的作用下轉(zhuǎn)化成zui終的黃色。顏色的深淺和樣品中的待測因子呈正相關(guān)。
