ELISA常用的方法主要包括雙抗體夾心法、間接法、競爭法以及BAS-ELISA。
（1）大鼠*結(jié)合蛋白ELISA試劑盒，RBP ELISA Kit  雙抗體夾心法是檢測抗原zui常用的方法。夾心法利用兩種一抗對目標(biāo)抗原進(jìn)行捕獲和固定，在確保靈敏度的同時大大提高了反應(yīng)的特異性，該方法的檢測靈敏度可提高到pg級的水平。將已知抗體包被到固相表面，加入待檢標(biāo)本，標(biāo)本中若含有相應(yīng)抗原即與固相表面的抗體結(jié)合，洗滌去除未結(jié)合成分，加入該抗原特異的酶標(biāo)記抗體，洗去未結(jié)合的酶標(biāo)記抗體，加底物后顯色。若標(biāo)本中無相應(yīng)抗原，固相表面即無抗原結(jié)合，加入的酶標(biāo)記抗體則不能結(jié)合于固相并被洗滌去除，當(dāng)加入無色底物后，因無酶催化故不顯色。雙抗體夾心法適用于測定2價或2價以上的大分子抗原，但不適用于測定半抗原及小分子單價抗原，因其不能形成兩位點夾心。
（2）間接法是檢測抗體zui常用的方法，其原理為利用酶標(biāo)記的抗抗體以檢測已與固相結(jié)合的受檢抗體，故稱為間接法。先將待測的蛋白包被在孔板內(nèi)，然后依次加入一抗、酶標(biāo)記u akn 的二抗和底物顯色，通過儀器（例如酶標(biāo)儀）定量檢測抗原。這種方法操作簡單但由于高背景而特異性較差，目前已逐漸被夾心法取代。間接法的優(yōu)點是只要變換包被抗原就可利用同一酶標(biāo)二抗建立檢測相應(yīng)抗體的方法。間接法成功的關(guān)鍵在于抗原的純度。間接法中另一種干擾因素為正常血清中所含的高濃度的非特異性抗體。
（3）競爭法可用于測定抗原，也可用于測定抗體。當(dāng)抗原材料中的干擾物質(zhì)不易除去，或不易得到足夠的純化抗原時，可用此法檢測特異性抗體。其原理為標(biāo)本中的抗體一定量的酶標(biāo)抗體競爭與固相抗原結(jié)合。標(biāo)本中抗體量越多，結(jié)合在固相上的酶標(biāo)抗體愈少，因此陽性反應(yīng)呈色淺于陰性反應(yīng)。如抗原為高純度的，可直接包被于固相。如抗原中有干擾物質(zhì)，直接包被不易成功，可采用捕獲包被法，即先包被與固相抗原相應(yīng)的抗體，然后加入抗原，形成固相抗原。洗滌除去抗原中的雜質(zhì)，然后再加入標(biāo)本和酶標(biāo)抗體進(jìn)行競爭結(jié)合反應(yīng)。小分子抗原或半抗原因缺乏可用于夾心法的兩個以上的位點，因此不能用雙抗體夾心法進(jìn)行測定，可以采用競爭法模式。其原理是標(biāo)本中的抗原和一定量的酶標(biāo)抗原競爭與固相抗體結(jié)合。標(biāo)本中抗原量含量愈多，結(jié)合在固相上的酶標(biāo)抗原愈少，zui后的顯色也愈淺。
