酶聯(lián)免疫技術(shù)在科學(xué)實驗的應(yīng)用：
1）酶聯(lián)免疫組織化學(xué)技術(shù)用于組織細胞上抗原、抗體成分的定位。
2 測定法用來檢測液體中可溶性的抗原、抗體成分。
3）酶聯(lián)免疫技術(shù)中的競爭法是測定小分子抗原的方法。尤其適用于食品衛(wèi)生分析。食品衛(wèi)生分析中所需測定的物質(zhì)大都是低分子質(zhì)量的，但免疫動物產(chǎn)生抗體的抗原其相對分子質(zhì)量要求必須很大(至少50 000)，因此，首先必須將小分子物質(zhì)結(jié)合上高分子蛋白質(zhì)，才能作免疫之用，而為了能使小分子物質(zhì)結(jié)合上高分子蛋白質(zhì)，則必須在小分子物質(zhì)上導(dǎo)人一個活性基團，同時直接競爭法所需的酶標抗原的制備也需首先在小分子物質(zhì)上導(dǎo)人一個活性基團。
由于該化學(xué)修飾反應(yīng)有一定的技術(shù)難度，有關(guān)專家就采用了間接競爭法來改進測定步驟，用酶標抗體代替了酶標抗原，因為酶標抗體的制備技術(shù)相對比較成熟，并有現(xiàn)成的商品，然而這卻使整個實驗操作過程不如直接競爭法那么簡便快捷。
競爭酶聯(lián)免疫吸附分析法有特異性強、干擾小、樣品的預(yù)處理簡便、快速、高效的特點，并且對設(shè)備裝置的投資較少。
定性測定結(jié)果判定中常用的一些縮寫：
(1)S／CO：其中S為sample(樣本)或specimen(標本)的簡寫，表示的是標本測定的吸光度值，CO為cut-off值的簡寫。除競爭抑制法外，其他ELISA定性測定模式中，當S／CO值大于或等于1時，標本的測定為陽性，小于1時為陰性。
(2)S／N或P／N：其中S同(1)，N為negative(陰性對照)的簡寫，P為patient(患者)的簡寫。較早的試劑盒很多都使用S／N或P／N≥2．1為陽性判定標準，現(xiàn)仍有一些試劑盒使用這種方式。這種方式與S／CO方式無根本性區(qū)別，只不過是前者將陰性對照(N)的2．1倍視為cut-off值而已。
