ELISA試劑盒的包被條件與封閉：包被用抗原或抗體的濃度，包被的溫度和時(shí)間，包被液的pH等應(yīng)根據(jù)試驗(yàn)的特點(diǎn)和材料的性質(zhì)而選定?？贵w和蛋白質(zhì)抗原一般采用pH9.6的碳酸鹽緩沖液作為稀釋液，也有用pH7.2的磷酸鹽緩沖液及pH7~8的Tris-HCL緩沖液作為稀釋液的。通常在ELISA板孔中加入包被液后，在4-8℃冰箱中放置過(guò)夜，37℃中保溫2小時(shí)被認(rèn)為具有同等的包被效果。包被的zui適當(dāng)濃度隨載體和包被物的性質(zhì)可有很大的變化，每批材料需通過(guò)實(shí)驗(yàn)與酶結(jié)合物的濃度協(xié)調(diào)選定。一般蛋白質(zhì)的包被濃度為100ng/ml-20ug/ml。
封閉(blocking)是繼包被之后用高濃度的無(wú)關(guān)蛋白質(zhì)溶液再包被的過(guò)程。抗原或抗體包被時(shí)所用的濃度較低，吸收后固相載體表面尚有未被占據(jù)的空隙，封閉就是讓大量不相關(guān)的蛋白質(zhì)充填這些空隙，從而排斥在ELISA其后的步驟中干擾物質(zhì)的再吸附。封閉的手續(xù)與包被相類似。zui常用的封閉劑是0.05%-0.5%的牛血清白蛋白，也有用10%的小牛血清或1%明膠作為封閉劑的。脫脂奶粉也是一種良好的封閉劑，其zui大的特點(diǎn)是價(jià)廉，可以高濃度使用（5%）。高質(zhì)量的速溶食用低脂奶粉即可直接當(dāng)作封閉劑使用，但由于奶粉的成份復(fù)雜，而且封閉后的載體不易*保存，因此在試劑盒的制備中較少應(yīng)用。 
封閉是否必要，取決于ELISA的模式及具體的實(shí)驗(yàn)條件。并非所有的ELISA固相均需封閉，封閉不當(dāng)反而會(huì)使陰性本底增高。一般說(shuō)來(lái)，雙抗體夾心法，只要酶標(biāo)記物是高活性的，操作時(shí)洗滌*，不經(jīng)封閉也可得到滿意的結(jié)果。特別是用單抗腹水直接包被時(shí)，因其中大量非抗體蛋白在包被時(shí)同樣也吸附在固相表面，業(yè)已起到了類似封閉劑的作用。但在間接法測(cè)定中，封閉一般是不可少的。包被好的ELISA板干燥后放入密封袋或錫袋中，在低溫可保存數(shù)月。
