1 選擇試劑：選擇質(zhì)量優(yōu)良的檢測試劑，嚴(yán)格按照試劑說明書進(jìn)行操作，操作前將試劑在室溫下平衡30-60分鐘。
2 加樣：可能原因： 
1）血清或血漿標(biāo)本分離不好即進(jìn)行加樣； 
2）手工操作中，加樣板過多造成加樣后放入孵箱前等待時(shí)間過長(特別是室內(nèi)溫度較高時(shí))； 3）加完標(biāo)本再加酶試劑時(shí)酶濺出孔外。
解決辦法： 
1） 標(biāo)本為血清：將血液先自然存放1-2小時(shí)后，再用3000rmp離心15分鐘；標(biāo)本為血漿：必須使用含抗凝劑的血液標(biāo)本收集管，采血后必須立即顛倒*混合5-10次，放置一段時(shí)間后，3000rpm離心15分鐘；若在幾天內(nèi)檢測，可放在2-8℃冰箱中，若要貯存，則置于-20℃的低溫冰箱內(nèi)。 
2) 加樣后及時(shí)放入孵箱。 
3) 加酶試劑后用吸水紙?jiān)诿笜?biāo)板表面輕拭吸干。 
4) 如果采用AT或其他全自動(dòng)加樣，選擇FAME或其他后處理儀器加酶試劑。 
5) 標(biāo)本較多時(shí)，請分批操作。
3 孵育：可能原因： 
1) 孵育時(shí)未貼封片或加蓋，使標(biāo)本或稀釋液蒸發(fā)，吸附于孔壁，難于清洗*； 
2) 孵育時(shí)間人為延長，導(dǎo)致非特異性結(jié)合緊附于反應(yīng)孔周圍，難以清洗*。解決辦法： 
1) 貼封片或加蓋； 
2) 按說明步驟嚴(yán)格控制操作時(shí)間。
4 洗板：可能原因： 
1) 采用手工洗板,孔與孔之間液體交叉。 
2) 采用半自動(dòng)洗板機(jī)洗板時(shí)，洗液量不足，導(dǎo)致洗板不*；洗板針堵塞，抽吸不*；洗板不暢，導(dǎo)致洗板效果差。 
3) 反應(yīng)板過多造成洗板等待時(shí)間長。 解決辦法： 
1) 保證洗液注滿各孔，洗板針暢通，洗完板后在吸水紙（選擇干凈、無或少塵的吸水材料）上輕輕拍干； 
2) 合理安排，或多用幾臺(tái)洗板機(jī)。

1) 顯色劑配制后放置時(shí)間過長或使用過期顯色劑； 
2) 加顯色劑時(shí)濺出孔外造成液體回流。解決辦法： 
1) 顯色劑盡量在臨用前配制，堅(jiān)持不用過期顯色劑，肉眼可見淺藍(lán)色的TMB顯色劑不用； 2) 加樣時(shí)保持顯色劑不外流； 
3) A、B液應(yīng)避免接觸金屬器械。
6 終止：可能原因如加終止液時(shí)產(chǎn)生較多氣泡，導(dǎo)致假陽性增加。所以在加終止液時(shí)應(yīng)避免產(chǎn)生氣泡。
