ELISA的基礎(chǔ)是抗原或抗體的固相化及抗原或抗體的酶標(biāo)記。在測定時(shí)，受檢標(biāo)本（測定其中的抗體或抗原）與固相載體表面的抗原或抗體起反應(yīng)。此時(shí)固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例。再加入酶標(biāo)記的抗原或抗體，也通過反應(yīng)而結(jié)合在固相載體上。結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學(xué)活性，酶標(biāo)記的抗原或抗體既留存其免疫學(xué)活性，又留存酶的活性。加入酶反應(yīng)的底物后，底物被酶催化成為有色產(chǎn)物，產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān)，故可根據(jù)呈色的深淺進(jìn)行定性或定量分析。因?yàn)槊傅拇呋屎芨?，間接地放大了免疫反應(yīng)的結(jié)果，使測定方法達(dá)到很高的敏感度。
