人β淀粉樣蛋白1-40(Aβ1-40)ELISA試劑盒原理的簡要說明書：Aβ1-40，Aβ 主要包括Aβ 40 和Aβ 42，其中Aβ 42 更容易聚合，也更具有致病性。Aβ 的產(chǎn)生需要有β- 和γ- 分泌酶的活性存在。APP 的胞外區(qū)首先被β- 分泌酶在Aβ 的1 或11 氨基酸位置水解，所產(chǎn)生的跨膜片段APP βCTF 再進(jìn)一步被γ- 分泌酶在膜雙分子層內(nèi)Aβ 的40 或42位置水解，從而生成Aβ40 或Aβ42。決定Aβ 產(chǎn)生的γ- 分泌酶是一個(gè)高分子量的復(fù)合體。
