包被用人NOGO-A抗體（Nogo-A Ab）ELISA試劑盒抗原：用于包被固相載體的抗原按其來源不同可分為天然抗原、重組抗原和合成多肽抗原三大類。天然抗原可取自動物組織、微生物培養(yǎng)物等，須經(jīng)提取純化才能作包被用。
包被用抗體：包被固相載體的抗體應(yīng)具有高親和力和高特異性，可取材于抗血清或含單克隆抗體的腹水或培養(yǎng)液。如免疫用抗原中含有雜質(zhì)（即便是極微量的），在抗血清中將出現(xiàn)雜抗體，必須除去（可用吸收法）后才能用于ELISA，以保證試驗的特異性。抗血清不能直接用于包被，應(yīng)先提取IgG，通常采用硫酸銨鹽析和Sephadex凝膠過濾法。一般經(jīng)硫酸銨鹽析粗提的IgG已可用于包被，高度純化的IgG性質(zhì)不穩(wěn)定。如需用高親和力的抗體包被以提高試驗的敏感性，則可采用親和層析法以除去抗血清中含量較多的非特異性IgG。腹水中單抗的濃度較高，特異性亦較強，因此不需要作吸收和親和層析處理，一般可將腹水作適當(dāng)稀釋后直接包被，必要時也可用純化的IgG。應(yīng)用單抗包被時應(yīng)注意，一種單抗僅針對一種抗原決定簇，在某些情況下，用多種單抗混合包被，可取得更好的效果。
