【產(chǎn)品說明書】
【原理】
流感病毒檢測卡以雙抗體夾心法為基礎(chǔ),采用免疫層析金標記技術(shù),快速檢測流感病毒。
【試劑組成】
1. 流感病毒快速檢測卡40片/盒
2. 樣本稀釋液40管/盒
3. 棉簽40支/盒
【操作方法】 一、樣品制備:
1. 本檢測卡采集樣品為眼、氣管分泌物。將棉簽插入分泌物zui多的部位,輕輕搖動棉簽,讓棉簽充分吸收分泌物。
2. 將棉簽在稀釋液中充分攪拌并反復擠壓試管壁,讓分泌物充分溶解到稀釋液中,得到待檢樣品。
3. 樣品一般須當即進行檢測,否則應(yīng)冷藏保存,超過24小時的,應(yīng)該冷凍保存。
二、操作步驟:
1. 使用前將試劑盒和樣品恢復至室溫。
2. 將棉簽浸入裝有樣品稀釋液的試管,充分攪拌混勻后,用一次性滴管取上清液。
3. 取出檢測卡,開封后平放于桌面上,從滴管中緩慢而準確地逐滴加入2–3滴混合液。
4. 加樣品液后,約30秒內(nèi),紅色的液體從靠樣品孔的觀察窗邊緣涌出。朝另一方向流動。
5. 五分鐘后判斷結(jié)果,超過三十分鐘的結(jié)果判讀無效。
【結(jié)果判定】
1. 陽性:在觀察孔內(nèi),若對照線顯色,檢測線同時顯色,判為陽性(如圖一、圖二)。陽性結(jié)果表明檢測液
中含有流感病毒。
2. 陰性:在觀察孔內(nèi),若對照線顯色,而檢測線不顯色,判為陰性(如圖三)。陰性結(jié)果表明檢測液中不含流感病毒。
3. 無效:在觀察孔內(nèi),若對照線不顯色,則結(jié)果無效,建議重新測試(如圖四、圖五)。
【注意事項】
1. 本品如果購買時發(fā)現(xiàn)過期、破損、污染、無效的產(chǎn)品,請在購買處進行更換。
2. 測試樣品來自動物,可能有潛在感染性,樣品和使用過的試劑應(yīng)被看作微生物危險品處理。
3. 所有檢測卡啟封后馬上使用,此前不要隨意打開。
4. 本品為一次性產(chǎn)品,請勿二次使用;請勿使用非本品隨附的稀釋液。
【儲存及有效期】
1. 保存于干燥陰涼處(2~30℃),有效期為18個月,生產(chǎn)日期見外包裝盒。
2. 打開包裝后,在1小時內(nèi)使用。

存在環(huán)境核酸檢測試劑盒
軍團菌是隱藏在空調(diào)制冷裝置中的致病菌,隨冷風吹出浮游在空氣中,吸入人體后會出現(xiàn)上呼吸道感染及發(fā)熱的癥狀,嚴重者可導致呼吸衰竭和腎衰竭。軍團菌zui早是由于*室內(nèi)密閉作業(yè),導致群體發(fā)生上呼吸道感染而得名。軍團病是一種非常嚴重的、有時可以致命的肺炎。軍團病是由軍團桿菌引起,這種細菌產(chǎn)生在自然環(huán)境中,在溫水里及潮熱的地方蔓延。人工供水系統(tǒng)有時也能為軍團桿菌的大量繁殖提供生存環(huán)境。這些系統(tǒng)包括淋浴器、礦泉池、噴泉以及空調(diào)設(shè)備的冷卻水塔。人們通常是由于呼吸了被軍團桿菌污染的水源散發(fā)的水霧而傳染上軍團病的。
鑒定方法核酸檢測試劑盒
1. 細菌培養(yǎng)法
軍團菌為革蘭氏陰性桿菌,專性需氧,胞內(nèi)寄生菌。軍團菌zui初是從以軍團菌病人肺組織感染的豚鼠中分離出來的。這種方法雖然有較高的可靠性,但非常昂貴,而且費時費力,不久就被平板培養(yǎng)法所取代核酸檢測試劑盒 。目前標準培養(yǎng)基為活性碳酵母浸膏瓊脂平板,也稱軍團菌生長平板(BCYE)。接種在 BCYE 平板上的樣品在溫度為35-37oC,培養(yǎng)10天,在濃度為2.5% CO2 的環(huán)境下培養(yǎng)更有利于軍團菌的生長。軍團菌的菌落通常呈白色、灰色、有熒光。軍團菌在 BCYE 平板上生長而在平板和半光氨酸缺失 BCYE-Cys 平板上不生長。
2.血清學玻片凝集法
軍團菌感染l周左右可檢測出血清中軍團菌特異性IgM抗體,2周左右可檢測到特異性IgG抗體。
現(xiàn)已發(fā)現(xiàn)嗜肺軍團菌有15個血清型。其中,嗜肺軍團菌血清1型(Lp1)與人類疾病關(guān)系zui密切,其次為血清4型(Lp4)和6型(Lp6)。
3.核酸擴增法
核酸擴增技術(shù),即聚合酶鏈式反應(yīng)(polymerase chainreaction,PCR),其基本原理是設(shè)計、合成兩條寡核苷酸,作為引物,對應(yīng)于待測病原微生物某一段特異性序列的兩端,然后在體外模擬DNA體內(nèi)復制的過程反復擴增,使靶序列放大上萬倍甚至上百萬倍而被檢測出來。
常規(guī)PCR:鑒定標準是通過電泳來判斷是否有擴增的核酸片段以及擴增產(chǎn)物的大小是否正確。
實時熒光PCR:通過對實時熒光PCR反應(yīng)的每一個循環(huán)產(chǎn)物進行熒光信號的實時監(jiān)測來判斷分析。
殺滅方法核酸檢測試劑盒
溫度殺菌:水加熱三分鐘到至少70°。
化學殺菌:大多用氯,二氧化氯,氯化鈉,次氯酸鈣溶劑殺菌,在使用化學品之前必須先清洗水管。
紫外線殺菌:照射紫外線短波,波長254納米可以使病菌無法活躍,但可能存在于變形蟲里。為更有力的除菌可以在附加使用超聲波。
薄膜技術(shù):越來越多的使用薄膜來去除微生物。一般可以利用薄膜的微孔讓直徑小于0.2微米的顆粒包括細菌,甚至部分病毒過濾掉。
診斷與治療核酸檢測試劑盒
軍團菌病是由軍團菌科細菌所致的肺部感染。多散在發(fā)病或小流行,亦可暴發(fā)流行。由空調(diào)、供水系統(tǒng)、霧化吸入污染的水源引起感染。本病特征為肺炎伴全身性毒血癥癥狀,嚴重者出現(xiàn)周圍回圈衰竭、呼吸衰竭。PCR檢測試劑盒X線表現(xiàn)在早期為單葉斑片狀陰影,繼而肺實變,病變迅速發(fā)展至多肺葉段,可有肺膿腫形成或少量胸腔積液征。確診依賴于特異性抗體的檢查及在痰、胸腔積液、肺組織活檢標本中分離出細菌。紅霉素、利福平對本病的治療效果較好。
臨床表現(xiàn)
1.緩慢起病,反復寒戰(zhàn)、高熱、伴頭痛、肌痛、乏力,常有腹痛、嘔吐、腹瀉和全身衰竭,多數(shù)病人有咳嗽、胸痛,少量粘液痰,可有血痰。嚴重患者有呼吸困難,紫紺,甚至呼吸衰竭,神經(jīng)精神癥狀,譫妄,定向力障礙等。
2.急性病容,呼吸急促。肺部有羅音和實變體征。約20%有相對緩脈。少數(shù)患者有胸膜摩擦音。
診斷依據(jù)
1.可為流行或散發(fā)發(fā)病,多發(fā)生于中老年或有慢性疾病患者。
2.發(fā)熱、大汗、咳嗽,咳白粘痰,伴胸痛、肌痛及乏力。神經(jīng)、精神癥狀或PCR檢測試劑盒、嘔吐、腹瀉,嚴重者可出現(xiàn)腎功能衰竭表現(xiàn)。
3.呼吸困難,相對緩脈。肺部可有濕羅音和實變體征或有胸膜摩擦音。
4.白細胞總數(shù)增高,肝功能可異常,血尿素氮增高。
5.PCR檢測試劑盒X線顯示單葉或多葉變化較迅速的片狀陰影,常伴有膿腫形成,胸腔積液征。
6.特殊檢查:直接熒光抗體試驗陽性。肺活檢組織用特殊染色軍團菌陽性; 間接熒光抗體試驗陽性。特異性血清抗體滴定>1:256,或雙份血清對比高4倍;放射免疫測定法或梅聯(lián)免疫吸附試驗測定尿中可溶性細菌抗原,痰、胸液和肺組織特殊培養(yǎng)有嗜肺軍團菌生長。
治療原則
1.抗菌療法。
2.對癥治療。
3.支持療法。

Legionnaires' disease is caused by Legionella bacteria, a bacteria that occurs in the natural environment and spreads in warm water and hot flashes. Artificial water systems can sometimes provide a living environment for the mass production of Legionella bacteria. These systems include showers, mineral pools, fountains and cooling towers for air-conditioning equipment. Legionella is Gram-negative bacilli, obligate aerobic, intracellular parasites. Legionella was initially isolated from guinea pigs infected with lung tissue of Legionnaires' patients.
