人B細(xì)胞分化因子(BCDF)ELISA試劑盒說(shuō)明書(shū)注意事項(xiàng)：
1．邊緣效應(yīng) 使用96孔板的ELISA測(cè)定中，常發(fā)現(xiàn)有“邊緣效應(yīng)"，即外周孔顯色較中心孔深。經(jīng)研究證實(shí)在溫育中的熱力學(xué)梯度可能是根本原因之所。聚苯乙烯本身為不良熱導(dǎo)體，在實(shí)驗(yàn)室的常規(guī)ELISA測(cè)定中，將板從室溫（通常在25℃左右）置于37℃溫箱，板也升溫時(shí)，在外周孔與中心孔之間可能存在一熱力學(xué)梯度。因此使用水浴或在將反應(yīng)溶液加入至板孔中時(shí)，將板和溶液均加熱至溫育溫度（如37℃），就可以很容易地排除“邊緣效應(yīng)"，并且可提高測(cè)定的重復(fù)性。 
2．加樣在現(xiàn)在的ELISA商品試劑盒中，必然有使用微量加樣器加入樣本的步驟。注意的關(guān)鍵點(diǎn)是：加樣不可太快，要避免加在孔壁上部，不可濺出和產(chǎn)生氣泡。加樣太快，無(wú)法保證微量加樣的準(zhǔn)確性和均一性。加在孔壁上部的非包被區(qū)，易導(dǎo)致非特異吸附。濺出會(huì)對(duì)鄰近孔產(chǎn)生污染。出現(xiàn)氣泡則反應(yīng)液界面有差異。所以，有時(shí)候一份標(biāo)本用相同的試劑盒這次測(cè)定為陽(yáng)性，下次測(cè)定為陰性，往往就是上述加樣及試劑的錯(cuò)誤所致。
